Latrophilin-1 (LPHN1), also known as adhesion G protein-coupled receptor L1 (ADGRL1), is encoded by the gene mapped to human chromosome 19p13.12. It belongs to the unique class of G protein-coupled receptors.

Latrophilin-1 (LPHN1) binds to black widow spider venom component α-latrotoxin (α-LTX) and promotes a surge of vesicular release from synaptic terminals and neuroendocrine cells through formation of membrane pores. LPHN1 participates in presynaptic calcium homeostasis by interaction with a teneurin ligand. In addition, it also facilitates trans-cellular adhesion through interaction with neurexins 1β and 2β Mutation or overexpression of LPHN1 leads to asthmatic airway smooth muscle cells (ASMC), suggesting its role in airway function.
