The NF-κB transcription factors are presented inactive in the cytoplasm by inhibitor protein IκB. In response to stimuli such as UV light or inflammatory cytokines, phosphorylation and subsequent degradation of IκB, allows for nuclear translocation of NF-κ B where it functions as transcription factor. IκB is phosphorylated by a serine kinase, IκB kinase (IKK). The IKK consists of three subunits, IKKα and IKKβ are the catalytic, IKKγ is a regulatory subunit. IKKα and its upstream kinase, NIK are the key kinases in the activation of the alternate or the non-canonical NFκB pathway. IKKα has been reported to function independent of either IKKβ or IKKγ in activation of NF-κB. IKKα also shows functions other than activation of NF-κ B. It binds the promoters of estrogen-responsive genes, Cyclin D1 and c-myc
Anti-IKKα?detects IKKα?(85 kDa) and has no cross-reactivity with IKKβ?or IKKγ.
