EGFR pathway substrate 15 (Eps15) is an endocytic adaptor protein. It contains three N-terminal conserved EH (epsl5 homology) protein-protein interaction domains, a central coiled-coil region, and a C-terminal domain containing the adapter protein (AP)-2 and AP-1 binding domains. The C-terminal domain also contains two ubiquitin interaction motifs (UIMs), required for Eps15 binding to ubiquitin. Eps15 colocalizes with markers of the plasma membrane clathrin-coated pits and vesicles.

Epidermal growth factor (EGFR) pathway substrate 15 (Eps15), that plays a central role in the clathrin mediated internalization and degradation of EGFR. Eps15 is required in clathrin mediated endocytosis including of synaptic vesicles. This protein has been shown to regulate the endosomal trafficking of EGFR, possibly through the recruitment of the ubiquitinated receptors to the rims of clathrin-coated pits. The EPS15 gene yields two isoforms that are believed to reside in distinct subcellular locations and thus implicated in different aspects of endosomal trafficking.
