EPS15 gene encodes a protein, which is a tyrosine kinase substrate and is expressed ubiquitously. It is mapped to chromosome 1p31-p32 with three N-terminal conserved EH protein-protein interaction domains, a central coiled-coil region, and a C-terminal domain containing the AP-2 and AP-1 binding domains. The C-terminal domain also contains two ubiquitin interaction motifs (UIMs), required for Eps15 binding to ubiquitin.

The encoded protein is an adaptor protein that plays a pivotal role in the clathrin mediated internalization and degradation of EGFR (epidermal growth factor receptor). It also regulates the endosomal trafficking of EGFR, possibly through the recruitment of the ubiquitinated receptors to the rims of clathrin-coated pits. Additionally, Dab2 recruits EPS15 protein and facilitates the internalization of integrin β1.
