HiCrome^™ 麦康凯山梨醇琼脂建议用于选择性分离食物和动物饲料中的大肠杆菌 0157:H7。本品按照 Rappaport 和 Henigh 法配制。该培养基含山梨醇而不含乳糖，建议用于检测 大肠杆菌 0157:H7 的肠致病性菌株，这些菌株可发酵乳糖但不发酵山梨醇，因而会产生无色的菌落。大肠杆菌的山梨醇发酵菌株则产生粉红色菌落。变为红色的原因是：山梨醇生成酸、吸收中性红以及随后当培养基 pH 值低于 6.8 时染料发生颜色变化。大肠杆菌 0157:H7 被认为是出血性结肠炎的起因。D加入生化级指示剂用来检测对大肠杆菌呈特异性的 β-D-葡萄糖醛酸苷酶。具有 β-D-葡萄糖醛酸苷酶的大肠杆菌菌株在培养基上表现为蓝色菌落。而大肠杆菌 0157 的肠致病性菌株无 β-D-葡萄糖醛酸苷酶活性，因此产生无色的菌落。可发酵山梨醇并具有 β-D-葡萄糖醛酸苷酶活性的大肠杆菌则产生紫色菌落。结晶紫和胆盐可抑制大部分的革兰氏阳性微生物。加入亚碲酸盐-头孢克肟添加剂可使培养基具有选择性。亚碲酸钾可从其他大肠杆菌血清型中选择血清型 0157，并抑制气单胞菌和普罗威登斯菌。头孢克肟则可抑制变形杆菌。

Suspend 50.1g in 1 litre distilled water. Boil gently to dissolve the medium completely. DO NOT AUTOCLAVE. Cool to 50℃. Mix well and pour into sterile petri plates. If desired add Tellurite-Cefixime Supplement to the molten and cooled medium (50℃) before pouring into sterile petri plates.
